Plot[]
Wilbur gets tired of living on the farm and wants to go on vacation. Him, Cardigan, and Charlotte's three daughters escape to Georgia, so they could take a vacation, while Fern and her family go looking for them. First, Wilbur and his friends go to Atlanta, then Macon, then finally in Savannah, where Fern would find them. Wilbur and his friends are safely returned home.

- Harrsion Chad as Cardigan, a young black lamb who returned from the Second movie.
- Amanda Bynes as Nellie, one of Charlotte's three daughters. She serves as their leader.
- Anndi McAfee as Joy, the cynical, sarcastic member of the trio.
- Maria Bamford as Aranea, the kinder, gentler member of the trio.
- Tom Kenny as Jeffrey, a young gosling who returned from the First movie. He is Wilbur's old friend now he is friends with both Wilbur and Cardigan and he is the son of Gwen.
- Jim Cummings as Mr. Zuckerman, the current owner of Wilbur and Cardigan.
- Dawnn Lewis as Bessie, a brown female cow owned by Mr. Hirsch. She is friends with Wilbur and Cardigan.
- Cree Summer as Katie, a young brown female calf and Bessie's daughter.
- Tiffany Haddish as Julie, a black and white female cow. She is friends with Wilbur, Cardigan and Bessie.
- Charlie Adler as Templeton, a care-free, egotistical rat who lives at Homer's farm.
- Laraine Newman as Gwen, she is the goose who Wilbur befriended in the First film. She is Wilbur's friend and Jeffrey's mother.
- Miranda Cosgrove as Fern Arable, the spirited young girl who saved Wilbur in the First film.
